Quantitative Phase Imaging for Histology

Label-free quantitative imaging for high-contrast tissue analysis

Photonis, Xenics and El-Mul are now unified under the Exosens brand. Read more →

Quantitative Phase Imaging (QPI) is a label-free microscopy technique that generates highly contrasted, artefact-free images of biological tissues without staining or reagents. Using Phasics' patented Quadriwave Lateral Shearing Interferometry (QWLSI) technology, it measures the Optical Path Difference (OPD) introduced by the tissue from a single acquisition, providing quantitative information related to tissue structure, constituent density and morphology. Easily integrated into existing optical microscopes as a plug-and-play camera, the Phasics solution enables fast, non-destructive tissue analysis and supports multimodal imaging as well as automated digital pathology workflows based on machine learning and artificial intelligence.

Quantitative phase imaging brings high contrast in histology

The Phasics solution delivers highly contrasted images of tissues without any staining or reagent. These images have no artefacts and help with observing structures in the tissue such as cells, fibers, and vessels composing the extra-cellular matrix. They also help identify tumorous areas in the tissue. As the technique is label-free, the protocol is fast and easy. This high contrast is generated using the local delay introduced in the light path by the tissue. This delay – also called phase shift or optical path difference (OPD) is proportional to the local constituent density.

  • Specific contrast for fibers: When using polarized light, specific contrast enhancement is offered for structures such as collagen in tissues or stress fibers in cells…
  • Quantification in thick tissues: Thanks to an advanced phase tomography technique based on an easy sample z-scan, valuable quantitative parameter measurements are made in thick tissues.
  • Easy integration to high-content screening platforms: valuable data for automated diagnosis using machine learning algorithms and artificial intelligence processes

The Phasics innovative technique enables fast protocols for tissue imaging with no staining nor labelling. The resulting images are artefact-free and offer high contrast to observe tissue structures including cells and fibers.

It can be applied to:

  • Patient clinical follow-up: biopsy, anatomic pathology including digital pathology and rapid intraoperative examinations
  • Cancer diagnosis and monitoring using collagen orientation in tumors
  • Fiber detection and identification
  • Tissue morphology study
